چکیده
The recently emerging stem-cell artificial niche engineering in induced pluripotent stem cell (iPSCs) 3D cultures has provided enormous opportunities to fully utilize the potential of these cells biomedical applications. Although a chemically defined environment can supply with desirable safety for clinical use, establishing an degradable controlled release proliferated under mild conditions is still big challenge. Here, advanced releasable iPSC reported based on dendritic polyglycerol and poly(N-isopropylacrylamide)-co-polyethylene glycol polymers via physical–chemical cogelation strategy. Benefiting from synthetic materials their precise cooperation by covalent cross-linking physical phase transition, cogelation-based system be adjusted optimal parameters owns high biocompatibility support robust production quality iPSCs excellent expansion efficiency. Moreover, expanded released out controllably only adjusting temperature. Overall, this hydrogel scaffold shows great promise culture downstream

Induced Pluripotent Stem Cells: Challenges and Opportunities
Regenerative capacity of mammals is limited and can rarely regenerate a specific organ or tissue fully. Due to these limitations, regenerative medicine seeks efficient and safe cell sources for regeneration of damaged tissues and organs or treatment for incurable diseases. Human embryonic stem cells (HESCs) hold two important properties called self renewal and pluripotency. However, the use of ...

To translate recent advances in induced pluripotent stem cell biology to clinical regenerative medicine therapies, new strategies to control the co-delivery of cells and growth factors are needed. Building on our previous work designing Mixing-Induced Two-Component Hydrogels (MITCHs) from engineered proteins, here we develop protein-polyethylene glycol (PEG) hybrid hydrogels, MITCH-PEG, which f...
متن کاملMucin-Inspired Thermoresponsive Synthetic Hydrogels Induce Stasis in Human Pluripotent Stem Cells and Human Embryos
Human pluripotent stem cells (hPSCs; both embryonic and induced pluripotent) rapidly proliferate in adherent culture to maintain their undifferentiated state. However, for mammals exhibiting delayed gestation (diapause), mucin-coated embryos can remain dormant for days or months in utero, with their constituent PSCs remaining pluripotent under these conditions. Here we report cellular stasis fo...
متن کاملLarge-Scale Expansion of Human Embryonic and Induced Pluripotent Stem Cells for Cell Therapy Applications
Successful isolation, derivation and culturing of human pluripotent stem cells, including human embryonic stem cells (hESCs) and human induced pluripotent stem (hiPSCs) cells in laboratory scale has opened new horizones for cell therapy applications such as tissue engineering and regenerative medicine. However, most of the cell therapy protocols using these unique cells require large number of ...
متن کاملHydrogels as feeder-free scaffolds for long-term self-renewal of mouse induced pluripotent stem cells.
Expanding undifferentiated induced pluripotent stem (iPS) cells in vitro is a basic requirement for application of iPS cells in both fundamental research and clinical regeneration. In this study, we intended to establish a simple, low cost and efficient method for the long-term self-renewal of mouse induced pluripotent stem (miPS) cells without using feeder-cells and adhesive proteins. Three sc...
